Hybrid droplet/digital microfluidic platform with integrated optical detection system for high-throughput genetic engineering

We have developed integrated circuits for manipulation of droplets that combine droplets-in-flow and digital (electrowetting-on-dielectric) formats for generation, merger, and sorting of droplets. The hybrid platform combines the high throughput offered by analog droplet microfluidics with the high level of control afforded by digital microfluidics. A novel fiber-optic-based droplet tracking system is devised for tracking and optical detection of droplets. The detector also serves as a trigger for sorting of droplets. The platform is useful for many high-throughput applications such as screening of enzymes and combinatorial DNA assembly and transformation.
